Lung cancer hasbecome the one of the major threats to world healthcare, with death rate of oneout of four deaths in both men and women. It is among one of reason of cancer related deaths all over the world1. Golbally lung cancer accounts forabout 13 % of new cancer cases and 19 % fatality . In India, lung cancer accounts for6.9 % of new cases and 9.3% deaths in both sexes, it is the commonestcancer and cause of cancer related mortality in men, and second most in women after breast andovarian carcinoma 2. Depending on tumour size and stages oflung cancer several therapeutic regimens are available to treat lung cancer.
Stilltraditional treatments like chemotherapy and radiation therapy are associatedwith several adverse effects on the human system. Even though of the progress in development of several anticancer drugs, chemoresistanceis main obstacle that results in the failure of treatment3.Multiple factors are involved in development of chemoresistance such asinactivation of chemotherapy treatment, variation in tumor suppressor gene,anti and pro apoptotic proteins. Understanding the moleculer role of tumorencoded gene in development of drug resistance has great significance indevelopment of novel therapeutic target and also progress in diagnosis4.Tissuetransglutaminase 2 (TG2) is one ofubiquitously expressed protein among 7 members of Tissue TG family. It is knownto be involved in tumor metastasis 5. It is distributed amongmajor cell types and tissues and plays a significant role in several biologicalprocesses.
It take part in post-translation modification transamidase activity it is involved inGTPase, ATPase, protein kinase, protein disulfide, and isomerase activity 6. Up-regulated TG2 levels leads to differentpathological disorders such as Alzheimer’s, Parkinson’s disease, multiplesclerosis, celiac along with tumourdevelopment while Over expression of TG2is associated with malignant effectsleading to tumorigenesis, invasion, cell differentiation, and failure in apoptosis7.As a consequence, TG2 is known to have a significant impact in lungcarcinoma, especially towards non-small cell lung carcinoma 8.Targeting TG2 protein may be valuable in identifying potential inhibitorsagainst lung cancer. Numerous natural compounds have been reported to have manybiological activities as antibacterial, antiviral and anti-cancer 9. Naturalproducts and their derivatives mimic over 50% of all the products that arebeing used. In the present study an attempt is made in silico investigation of natural inhibitors against TG2 fromplant origin.
The hit compounds obtained in this study could play an importantrole in designing personalized therapy against lung cancer patients.
